시설장비 설명 본 장비는 단백질을 크기 별로 분리하여 분석 할 수 있는 웨스턴 블랏(Western Blot)의 전체 과정을 자동화한 장비입니다. 최대 25개의 샘플을 3시간 이내에 측정 및 분석할 수 있습니다. 수작업으로 이루어지는 기존의 웨스턴 블랏 방식에서 발생할 수 있는 오차를 최소화함으로써 정확하고 재현성 있는 결과를 제공하고 실험결과 최종 획득까지 소요되는 시간을 획기적으로 단축시킬 수 있습니다. 또한, 기존 웨스턴과 달리 단백질을 멤브레인(membrane)으로 transfer하는 과정이 없어 단백질의 손실이 없고, 소량의 시료만으로도 분석이 가능합니다. 특수 물질이 코팅된 모세관(Capillary) 내부에서 단백질이 크기별로 분리된 후 특수물질에 의해 모세관에 고정됩니다. 이 후 특정 단백질에 1차, 2차 항체가 결합하고, 루미놀(luminol) 반응을 통해 화학적 발광 신호를 나타냅니다. 이 신호를 장비의 detector에서 기록하여 소프트웨어를 통해 자동으로 분석하여 결과를 확인할 수 있습니다. 밴드(band) 형태의 결과와 발광신호의 강도 피크, 정량값 등 다양한 분석 결과를 제공합니다. 특히, 정량값의 경우 기존 웨스턴 블랏과 달리 밴드에서 환산한 값이 아닌 발광신호의 강도를 수치화하였기 때문에 정확도가 월등히 높습니다.
구성 및 성능 1) Western Blot Automation System (1) It provides fully automated complete solution for protein detection and characterization which performs all the manual processes associated with a traditional Western blot. (2) Protein sizing, quantitation and immunodetection of proteins is performed in a nano-immunoassay system. (3) The system uses fluorescent and biotinylated labeled molecular weight size standards and proprietary software to calibrate each capillary and assign molecular weight sizing to detected proteins. (4) The immunoassay utilizes chemiluminescent detection and quantization of target proteins. (5) Sample and volume required: 0.6-1.2 μg, 3？5 μL/well or less.
